The Complement Factor H Related 5 (CFHR5) gene is speculated to have an important role in regulating fatty acid composition in sheep. The aim of this study was to investigate the polymorphism of the CFHR5 gene and its association with fatty acid composition and cholesterol of sheep in Indonesia. A total of 172 rams from seven Indonesian sheep breeds were used for this study. Identification of CFHR5 gene polymorphism was performed by Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR‑RFLP) using AciI restriction enzyme. The results showed that the CFHR5 gene in all sheep populations was polymorphic producing three genotypes, CC, CT and TT, respectively. The polymorphism had a significant effect (p<0.05) on fatty acid composition (miristoleic [C14:1], ginkgolic [C17:1], tridecanoic [C13:0], and heptadecanoic [C17:0] acids) and cholesterol. The breed and polymorphism had a significant effect (p<0.05) on fatty acid composition. The CC genotype is the preferred genotype and exhibits reduced levels of saturated fatty acids and cholesterol. The CFHR5 gene (SNP c.1011C>T) has the potential to be used as a genetic marker for selection of low saturated fatty acid composition and cholesterol in sheep.

1011 C>T dari gen CFHR5 pada domba yang dipelajari bersifat polimorfik dan memengaruhi komposisi asam lemak (miristoleik [C14.Genotip CC memiliki rasio PUFA/SFA tertinggi, kolesterol terendah, dan asam lemak jenuh terendah, sehingga dianggap baik untuk kesehatan.CFHR5 dapat digunakan sebagai penanda genetik untuk pemuliaan domba dengan komposisi lemak dan kolesterol yang menonjol.

Penelitian selanjutnya dapat mengeksplorasi interaksi antara CFHR5 dan gen lain yang terlibat dalam metabolisme lipid, seperti APOA5 dan HSD17B13, untuk menilai efek sinergis pada profil asam lemak turunan penyakit kardiovaskular; melakukan studi longitudinal pada populasi domba lokal dengan variasi lingkungan berbeda untuk menilai pengaruh interaksi gen‑lingkungan terhadap ketersediaan asam lemak rantai panjang dan kolesterol darah; serta mengaplikasikan teknik CRISPR‑Cas9 untuk menandai dan memodifikasi alel CFHR5 dalam garis jantan dan betina domba guna menilai dampak langsung perubahan alel terhadap kualitas daging dan komposisi lemak secara komprehensif.
